Role of TNF and LTA polymorphisms on biological markers of response to oxidative stimuli in coal miners: a model of gene- environment interaction
نویسندگان
چکیده
Introduction Interaction between genetic background and oxidative environmental stimuli in the pathogenesis of human lung disease has been largely unexplored. Methods A prospective epidemiologic study was undertaken in 253 coal miners. Intermediate quantitative phenotypes of response to oxidant exposure, including erythrocyte glutathione peroxidase (GSH-Px) and catalase activities were studied. Oxidant exposures studied were smoking habits and cumulative dust exposure assessed by job history and ambient measures. Disease phenotypes included subclinical computed tomography score at the first survey and X-ray profusion grades twice, 5 years apart, to assess established coal workers’ pneumoconiosis (CWP). Miners were genotyped for common functional polymorphisms in the gene for tumor necrosis factor α (TNF) and lymphotoxin α (LTA), two proinflammatory cytokines that have been implicated in the pathogenesis of chronic lung diseases. Results Regarding gene-environment interaction on intermediate phenotypes, results showed interaction of a promoter polymorphism at the –308 position in TNF with occupational exposure on erythrocyte GSH-Px activity with a significant association in those with high exposure (p = 0.003) whereas no association was observed among those with low exposure (interaction p = 0.06). Regarding gene-intermediate phenotype interaction on clinical outcome, results showed an association of CWP prevalence with a NcoI polymorphism in LTA in those with low catalase activity (p = 0.05) whereas no association was observed in those with high activity (interaction p = 0.03). No other significant association was observed. Conclusion Results suggest that interactions of genetic background with environmental exposure and intermediate response phenotypes are important components in the pathogenesis of CWP.

متن کاملThe Association between TNF-alpha Gene Polymorphisms and Endometriosis in An Iranian Population
Objective Objective: TNF-α is an important cytokine in acute inflammatory response to infective factors. It seems that this response increases in patients with endometriosis due to presence of cytokines such as TNF-α which has been investigated in patients with endometriosis in different populations. This study aimed at examining four polymorphisms; TNF-α -238G/A, -308G/A, -857C/T and -863C/A a...
متن کاملژنتیک رتینوپاتی دیابتی: بررسی نقش ژن VEGF
Background: VEGF is newly discovered growth factor that has diverse biologic properties. The bottom-line of these activities is conduction and orchestration of a series of reactions that are taking place at microvasculature of different tissues/organs. Among the growth factors, cytokines and other mediators that reflect meaningful alteration in their local/systemic level, VEGF is the distinct p...
متن کاملCB2 Agonist (AM1241) Improving Effect on Ovalbumin-Induced Asthma in Rats
Asthma is a disease characterized by spontaneous contraction of the airways in response to a wide variety of endogenous and exogenous stimuli. Many asthma models are used to mimic the human asthma model in the literature. In order to better understand the role of the cannabinoid (CB) 2 receptor in the ovalbumin (OVA)-induced asthma model, a combination of both selective CB2 agonist (AM1241) and...
متن کاملCooperativity in biological systems
Living organisms can sense and respond to external and internal stimuli. Response isdemonstrated in many forms including modulation of gene expression profiles, motility, secretion, cell death, etc. Nevertheless, all forms share a basic property: they depend on sensingsmall changes in the concentration of an effector molecule or subtle conformational changes ina protein and invoking the appropri...
